Entamoeba histolytica Culture Service
The life history of Entamoeba histolytica consists of an encapsulated phase and a trophozoite phase. Its infective phase is the mature encapsulation containing the tetranuclear nucleus. Infectious encysts in fecally contaminated food or drinking water cause infection through oral ingestion. Humans are suitable hosts for Entamoeba Histolytica, and cats, dogs and rats can also be occasional hosts. Its trophozoites can invade the intestinal mucosa, phagocytose red blood cells, destroy the intestinal wall, and cause ulceration of the intestinal wall; they can also enter other tissues or organs with the bloodstream and cause extraintestinal amebiasis.
In vitro cultures of Entamoeba histolytica are important in experimental diagnosis and preservation of the worm species, and the cultures are often fecal or abscess aspirates.

Robinson's medium is mainly used to culture not only the amoeba in lysate tissue, but also Entamoeba hartmani, Entamoeba coli grassi, Endolimax nana and other amoebas. We generally apply 6mL~7mL or smaller spiral covered culture tube.
We most commonly use BIS-33 medium (containing nutrient solution, vitamin mixture, adult bovine serum) as a liquid medium. Culture in 6ml glass covered culture tubes, as the amoeba is partly anaerobic metabolism, so the tube should be tightly covered and placed at an angle of 5°.
